局部应用骨保护素对鼠正畸牙移动影响的实验研究 被引量:5

摘要 目的探讨局部应用骨保护素(OPG)对牙移动影响。方法选用10周龄雄性SD(Spargue-Dewley)大鼠80只,实验组和对照组各40只。用40g力的拉簧牵引右上颌第1磨牙移向近中。将重组人骨保护素rhOPG-Fc和生理盐水分别注入实验组和对照组大鼠右上颌第1磨牙腭侧黏骨膜下。在1、4、8、12d分别处死实验大鼠,记录实验组与对照组右上颌第1磨牙近中的移动距离,观察右第1磨牙近中根压力侧牙周组织形态的变化和破骨细胞数量的变化。结果(1)实验组右上颌第1磨牙的移动距离在8d和12d时明显少于对照组(P<0.001)。(2)实验组大鼠右上颌第1磨牙近中根压力侧的牙槽骨骨吸收量少于对照组。(3)实验组大鼠右上颌第1磨牙近中根压力侧破骨细胞数量在4、8、12d时显著少于对照组(P<0.001)。结论局部注射OPG能抑制破骨细胞的分化,减少牙齿移动。 Objective To approach the effects of partial application of osteoprotegerin(OPG) to the orthodontic teeth movement in rats. Methods Eighty ten week-old SD(Spargue-Dewley) rats were tested. Each experiment group and control group contained 40 rats. The open load of 40g-spring drawed the right upper first molar into movement. In the experiment, the reformed fused prorein of humensr osteoprotegerin and physiological saline were injected into the palatine-side's mucoperiosteum of the right upper first molar of the experimental group and the control group respectively. Ten rats of each experiment group and control group were sacrificed on 1,4,8d and 12d. Then to record the moving distances of the right upper first molar of the experimental group and the control group respectively. The change of the periodental form and the quantity of the osteoclast were observed by histological see tion. Results (1)The moving distance of the right upper first molar of the experimental group on 8d and 12d was apparently lower than that of the control group(P〈0. 001). (2)The degree of the osteal absorbability of the alveolar bone in the pressure side of the mistal root of the right upper first molar in the experimental groups was lower than that in the control group. (3)The quantity of osteoclasts in the pressure side of the mistal root of the right first upper molar in the experimental groups varied on 4d,8d and 12d was obviously less than that in the control group(P〈0. 001). Conclusion Local delivery of OPG inhibits usteoclastogenesis and tooth movement.
