海南黎族冠心病与血管紧张素转化酶基因多态性的关系 被引量:1

A correlational study of angiotensin converting enzyme gene polymorphisms with coronary heart disease in hainan Li nationality

摘要 目的:研究海南黎族人群血管紧张素转化酶(ACE)基因多态性与冠心病(CHD)关系。方法:采用聚合酶链反应(PCR)技术,对海南黎族150例CHD患者(CHD组)和150例正常人(正常对照组)的ACE基因插入/缺失(I/D)多态性进行检测,观察DD、DI、II基因型频率及等位基因频率,并对所有普通PCR定为DD型的样本进行插入特异性PCR检测,以减少误分型率。结果:CHD组DD、DI、II基因的频率分别是24.7%、32.7%、42.6%;D及I等位基因频率分别为41.0%、59.6%;正常对照组DD、DI、II基因型频率分别为14.0%、44.0%、42.0%,D及I等位基因频率分别为36.0%、64.0%;2组之间的DD、DI、II基因频率及D、I等位基因频率,均差异有统计学意义(P<0.05)。结论:ACEI/D多态性与黎族CHD有显著关联,是海南黎族CHD的主要致病基因。早期应用ACE抑制剂、血管紧张素受体拮抗剂防治,有十分重要意义。 Objective: To study the correlation of the angiotensin converting enzyme (ACE) gene polymorphisms with coronary heart disease (CHD) in the Hainan Li nationality. Method:Used the polymerase chain reaction (PCR) to detect the polymorphisms of ACE gene insertion/deletion (I/D) in 150 patients with CHD and 150 healthy people from Hainan IA nationality. The genotype frequencies and allele frequencies of DD, DI and II were observed. Specific PCR detection was performed for patients who have been determined as DD by normal PCR to reduce misclassification rate. Result:In the C HD group of Hainan Li nationality, the genotype frequencies of DD, DI and II were 24.7%. 32.7%.42.6% respectively, and the allele frequencies of D and I were 41.0% and 59.0%. In the healthy control of Hainan Li nationality, the genotype frequencies of DD, DI and II were 14.0%,44.0%, 42.0% respectively, and the allele frequencies of D and I were 36.0% and 64.0%. There were significant differ ences both in the genotype frequencies of DD, DI and II, and in allele frequencies D and I between these two groups (P〈0.05). Conclusion:'Fbere were notable correlation between the ACE gene I/D polymorphisms and the CHD in Hainan Li nationality. The ACE gene polymorphisms are the major gene that causes CHD in Hainan Li nationality. It may be the independent risk factor in CHD in Hainan Li nationality. So it is very important to intervene with ACE inhibitors or angiotensin receptor blockers in the early stage of CHD.
