摘要
研究微量元素Mn、Fe、Zn对半夏组织培养诱导率、分化率的影响。分别以半夏无菌试管苗的叶片、叶柄、块茎为外植体,以MS+2,4-D 0.5 mg/L+6-BA 1 mg/L为基础培养基,改变其中Mn、Fe、Zn的浓度,每个因素设3个水平,正交试验设计,共选9个组合,每种外植体进行不同组合试验,寻找最佳的微量元素组合配方。结果表明:培养叶片叶柄的最佳微量元素浓度水平为Mn 5.5 mg/L,Fe为9.5 mg/L,Zn为2.0 mg/L;培养小块茎的最佳微量元素浓度水平为Mn7.5 mg/L,Fe 9.5 mg/L,Zn为1.0 mg/L。
To study the effect of Mn, Fe, Zn on the inductivity and tissue differentiation from different explants of Pinellia tissue culture. Leaves, petioles and tubers of Pinellia were cut and cultured on the MS+2,4-D 0. 5 mg/L+6- BA 1 mg/L medium with different concentration of Mn, Fe, Zn. The effects were studied by means of 3 factors and 3 levels orthogonal design experiments. The result indicated Mn 5.5 mg/L, Fe 9. 5 mg/L and Zn 2. 0 mg/L combine were advantageous to the culture of leaves and petioles; Mn 7. 5 mg/L, Fe 9. 5 mg/L and Zn 1.0 mg/L combine were advantageous to the tubers.
