中药枳术汤加味对大鼠肠黏膜屏障通透性的影响 被引量:1

Effect of the Zhishu Decoction on the permeability of rat intestinal mucosal barrier

摘要 [目的]观察中药枳术汤加味对大鼠肠黏膜屏障通透性的影响。[方法]60只雄性Wistar大鼠,随机分为3组,各20只。对照组,实验前给予0.85%氯化钠0.35 ml;模型对照(模型)组,术前给予0.85%氯化钠0.35 ml;模型治疗(治疗)组,术前给予中药枳术汤0.35 ml;均2次/d灌胃,连续7 d;通过夹闭肠系膜上动脉30 min后重新开放,建立肠道缺血-再灌注(IR)损伤模型;于灌注后1、24 h分别检测各组肠道细菌移位发生率、细菌培养阳性率、外周血浆D-乳酸浓度和二胺氧化酶(DAO)浓度。[结果]IR后1 h和24 h,比较肠道细菌移位阳性率、细菌培养阳性率、血浆D-乳酸和DAO浓度,模型组>对照组(P<0.05),治疗组<模型组(P<0.05)。[结论]中药枳术汤加味能抑制肠道细菌移位,降低肠黏膜的通透性,对肠黏膜屏障有较好的保护作用。 [Objective]To observe the effect of Chinese traditional medicine Zhishu Decoction on the permeability of rat intestinal mucosal barrier. [Methods]Sixty male Wistar rats were randomly divided into the control group (n=20),the model group (n=20) and the treatment group (n=20). Rats were continued to be given normal saline(0. 35ml, bid) for 7 days by using intragastric administration method,before experiment in control group or in model group, while rats were continued to be given the Zhishu Decoction(0.35ml, bid) for 7 days by using intragastric adminis- tration method before surgery in treatment group. The intestinal ischemia and reperfusion injury model had been established by blocking the upper mesenteric artery for 30 minutes and then re-opening it. The following indexes were measured after reperfusion for lh and 24h. incidence of intestinal bacterial translocation, the intestinal bacterial culture positive rate, peripheral serous concentration of D-lactate and the Diamine Oxidase. [Results]After ischemia-reperfusion for lh and 24h, the intestinal bacterial translocation incidence rate, bacterial culture positive rate, and serous concentration of D-lactate and Diamine Oxidase in model group were significantly higher than that in the control group (P〈0. 05) ; the incidence of intestinal bacterial translocation, the intestinal bacterial culture positive rate, peripheral serous concentration of D-lactate and the Diamine Oxidase in treatment group were markedly lower than those in model group (P〈0.05). [Conclusion] Chinese traditional medicine Zhishu Decoction can inhibit intestinal bacterial translocation, and reduce the permeability of the intestinal mucosa. So it can provide a better protection to the intesti-nal mucosal barrier function.
