摘要
目的:快速鉴定光叉叶委陵菜Potentilla bifurca var.glabrata中的酪氨酸酶抑制成分。方法:利用活性-高效液相色谱跟踪方法(bioassay-linked HPLC method),通过接收器接样,并用酪氨酸酶抑制强度,快速查找有效成分。结果:光叉叶委陵菜的甲醇提取物及其正丁醇萃取物显示较强的酪氨酸酶抑制作用。从光叉叶委陵菜的正丁醇萃取物中分离并鉴定一种黄酮类化合物,即槲皮素-4′-O-β-D-葡萄糖苷。与已知的酪氨酸酶抑制剂熊果苷(arbutin,IC50=0.28mmol.L-1)相比,槲皮素-4′-O-β-D-葡萄糖苷化合物具有更强的酪氨酸酶抑制作用。其IC50值为0.0019mmol.L-1。结论:该方法对中药及其复方药快速确定有效成分提供分析手段。
Objective: To identify the tyrosinase inhibitory constituent quickly from Potentilla bifurca. Method: The active constituent was found through fraction collecting and tyrosinase inhibitory activity by bioassay-linked HPLC method. Result: The meth- anol extracts and BuOH fraction of Potentilla bifurca showed strong tyrosinase inhibitory activities. From BuOH fraction of Potentilla bifurca, the tyrosinase inhibitory constituent was isolated and identified as flavonoid, quercetin-4'-O-β-D-glucoside. It express stronger tyrosinase inhibition than the known tyrosinase inhibitor, kojic acid (IC50 = 0. 28 mmol · L^-1) with ICso value of 0. 001 9 mmol · L^-1. Conclusion: Bioassay-linked HPLC fractionation method was provided for determination the active constituents quickly from herbal medicines.
