摘要
目的:采用HPLC研究"丹参饮"中君药丹参与使药檀香合用前后水溶性成分丹参素在家兔体内组织分布的变化。方法:家兔分别灌服丹参单煎液和丹参-檀香合煎液,于30,50,70min取心、脑、肝、肾,以10%的高氯酸为蛋白沉淀剂,醋酸乙酯为萃取剂,4-羟基苯甲酸为内标,测定君药丹参中丹参素在各组织中的含量。结果:丹参素在心、脑、肝、肾中的达峰时间均接近50min。丹参单煎和丹参、檀香合煎给药50min后,对应组织中丹参素的平均浓度分别为:C肾>C心>C脑>C肝和C肾>C肝>C脑>C心,合煎给药后,心、脑、肝、肾各组织中丹参素含量均大于丹参单煎给药时的含量。结论:"丹参饮"中使药檀香可显著提高丹参素在靶器官的含量,从而增强君药丹参对心脑血管疾病的治疗作用。
Objective: To investigate the influence of Shi herb (Santalum album, SA) to the tissue distribution of danshensu (DSS) which is the main hydrosoluble component of Jun herb (Salvia miltiorrhiza, SM) in rabbits by HPLC. Method: Rabbits were oral administrated decoction of SM and SM-SA, respectively. Perchloric acid ( 10% ) was used to precipitate the tissue samples of rabbits'heart, brain, liver, kidney, acetic ether was used to extracte supernataut, and the internal standard was p-hydroxybenzoic acid. The content of DSS of SM in tissues was detected. Result: The content of DSS reached the highest point close to 50 min in the men tioned tissues. Before and after co-administration, the sequences of average concentration of DSS in tissues were Ckidney 〉 Cheart 〉 Cbrain 〉 Cliver and Ckidney 〉 Cliver 〉 Cbrain 〉 Cheart respectively. Compared with SM administrated singly, the content of DSS in every tissues of co-administration was higher. Conlusion: In Danshenyin Formulae, SA can increase concentration of DSS in target tissues signifi- cantly, and therefore therapeutic effect of SM for cardiovascular disease is raised.
