黄芪注射液拮抗柴油机尾气颗粒物诱导的小鼠生殖功能改变 被引量:1

Antagonism effect of astragalus injection on the reproductive toxicity of mice exposed in Diesel exhaust particles

摘要 目的研究黄芪注射液对柴油机尾气颗粒物诱导的小鼠生殖功能改变的影响。方法5周龄雄性ICR小鼠60只随机分为对照PBS组,DEP染毒组(12、3、0.8μg/μl),黄芪注射液干预组(10mg/ml、20mg/ml)。染毒模型采用De Hennezel法。对照组、染毒组每日腹腔注射0.9%生理盐水1ml,干预组每日分别腹腔注射10mg/ml、20mg/ml的黄芪注射液1ml共28d,每周2次染毒连续4周,第6周处死。检测体重、睾丸、附睾脏器系数,精子密度、总活力,体外受精率。结果各组间睾丸重量、睾丸系数无统计学差异(P>0.05)。3、12μg/μl DEP染毒组与PBS组、0.8μg/μ1DEP染毒组比较附睾重量、附睾系数,精子密度、总活力、体外受精率均有统计学意义(P<0.05);3、12μg/μl DEP的黄芪注射液干预组与对应DEP染毒组比较附睾重量、附睾系数、精子密度、总活力、受精率有显著差异(P<0.05)但不存在剂量依赖关系(P>0.05)。总活力和体外受精率之间呈正相关(P=0.000)。结论每日连续运用10mg/ml、20mg/ ml的黄芪注射液1ml可以拮抗由柴油机尾气颗粒物诱导的雄性ICR小鼠生殖功能的改变。 Objective To study the antagonism effect of astragalus injection on the reproductive toxicity of mice exposed in diesel exhaust particles. Methods Adult male ICR mice with five weeks old were divided into PBS group, exposed Diesel Exhaust Particulates (DEP) group (12/3/0.8ug/ul) and astragalus injection interfered group (10mg/ ml, 20 mg/ml). The exposed model was established according to De Hennezel method. The test mice were exposed in diesel exhaust particulate twice weekly until four weeks, and the test mice were executed two week later Mice in PBS group and exposed diesel exhaust particulate groups were injected with lml 0.9%NaCI, and mice in interfered groups were respectively injected with 10mg/ml, 20 mg/ml astragalus injection everyday until the exposed test was over. The weight, relative testis/epididymis weights, sperm density, the total activity, and in vitro fertilization (IVF) rate of mice were comparatively analyzed. Results There were no statistical differences among the groups in testis weights, relative testis weight or weight (P〉0.05). There were statistical differences in epididymis weights relative ,epididymis weights sperm density, the total activity and IVF rate among 12 ug/ul and 3 ug/ul DEP-treated groups, PBS group and 0.8ug/ul DEP-treated groups (P〈0.05). Astragalus injection (10mg/ml, 20 mg/ml) could significantly improve epididymis weights relative, epididymis weights, sperm density, the total activity and IVF rate of exposed mice compared with that of 12/3ug/ul DEP group, but no dosedependent relationship was found for 10mg/ml/20 mg/ml astragalus injection and reproductive function of mouse. There existed a positive correlation between the total sperm activity and IVF rate (P=0.000). Conclusion The reproductive toxicity of exposed mice in DEP suspension can be ameliorated under 10mg/ml, 20mg/ml astragalus injection treatment each day.
