摘要
目的:探讨基质金属蛋白酶(MMP-9)siRNA对胃腺癌细胞系SGC7901中MMP-9表达的影响以及siRNA干扰后对SGC7901细胞侵袭和迁移的抑制作用。方法:设计合成针对MMP-9的siRNA,采用oligofectamine转染SGC7901胃腺癌细胞,分别采用实时定量聚合酶链反应(realtime PCR)和蛋白印迹法分别检测转染细胞后MMP-9mRNA及蛋白的表达。Matrgel 3D生长实验和Transwell实验检测细胞侵袭情况,Matrgel2D生长实验和划痕实验检测细胞迁移情况。结果:realtime PCR和蛋白印迹法显示转染MMP-9 siRNA后细胞中MMP-9mRNA和蛋白的表达较空白组和对照组明显降低。RNA干扰MMP-9基因后SGC7901细胞的侵袭和迁移能力也有明显下降。结论:靶向MMP-9的siRNA不仅能特异性的降低MMP-9mRNA及蛋白的表达,且能抑制人SGC7901胃腺癌细胞的侵袭和迁移。
Objective: To investigate the effect of MMP-9 siRNA on MMP-9 expression of human SGC7901 gastric adenocarcinoma cells and the inhibition effect on SGC7901 cell invasion and migration thereof. Methods: To design and synthesize siRNA targeting MMP-9, then transfect it into SGC7901 gastric adenocarcinoma cells with oligofectamine. The real-time PCR and Western blotting were used to observe the expression of MMP-9 mRNA and the protein. The Matrgel 3D growth experiment and transwell experiment were used to detect the invasion ability of cells. The Matrgel 2D growth experiment and scratch experiment were used to detect the migration ability of SGC7901 cells. Results: Real-time PCR and Western blotting assay indicated that the expressions of MMP-9 mRNA and the protein in SGC7901 ceils interfered by MMP-9 siRNA were significantly lower than those of other two groups. The invasion and migration ability of SGC7901 ceils were also degraded after siRNA interference of MMP-9 gene. Conclusion: The targeting RNA interference not only degraded the expression of MMP-9 mRNA and protein, but also inhibited the invasion and migration ability of SGC7901 cells.
