水稻叶片淀粉分解同源基因OsSEX1的转录表达

摘要 通过半定量RT-PCR分析了水稻的SEX1同源基因OsSEX1-1和OsSEX1-2的表达过程,结果表明:2个目的基因表达量均呈现昼夜节律变化。光照对2个基因的表达是必需的,其中以19:00时表达量最高,黑暗条件下表达量较低;叶片淀粉的含量也呈现昼夜节律变化;海藻糖抑制2个基因表达,并导致夜晚叶片淀粉异常积累;在自然光照/黑暗条件下,未观察到不同时段2个基因DNA甲基化差异。 The transcription of two SEX1 homologous(named OsSEX1-1 and OsSEX1-2)in rice leaves were analyzed with semi-quantitative RT-PCR method. Results showed that both genes were of circadian-expression character in rice leaf. Transcripts reached peak at 19:00 hut decreased at night. Light was essential for 2 genes' transcription. Starch contents in rice leaf also showed diurnal rhythm. In accordance with the excessive starch accumulation in rice leaf, neither OsSEXI-1 nor OsSEX1-2 expressed when trehalose was used as the only carbon source and no difference of DNA demethylation was detected either in OsSEX1-1 or in OsSEX1-2 at different diurnal stages. The result provided basis for the further study of expression and function of both genes in rice.
