摘要
为建立表达含前S基因乙肝病毒表面抗原的细胞株,将笔者所在实验室构建的表达含前S基因乙肝病毒表面抗原(HBsAg)的真核表达质粒共转染CHO/dhfr-细胞,通过筛选,获得可表达含前S基因乙肝病毒表面抗原的重组CHO细胞,然后进行亚克隆,用氨甲喋呤(MTX)加压选择,以ELISA法检测目的蛋白表达量,筛选获得稳定表达目的蛋白的细胞株,并对其进行特性鉴定。最终获得了C10和A10两株稳定高表达含前S基因乙肝病毒表面抗原的细胞株。
To establish efficient eukaryotic cell line to express hepatitis B surface antigen carrying pre-S, two kinds of eukaryotic expression vectors: pcHBSPP and psHBSPP were co-transfected into CHO/dhfr-cells. The transfected cells were selected by G-418 and MTX. The expression of target protein was detected by ELISA. The cell lines C10 and A10 stably expressing hepatitis B surface antigen carrying pre-S were established.
