摘要
目的探讨刃天青微孔板显色法在抗念珠菌药敏实验中的应用。方法采用NCCLS之M27-A方案测定伊曲康唑、氟康唑和两性霉素B对87株临床分离的念珠菌属真菌的最低抑菌浓度(MIC),终点判定同时采用NCCLS推荐的直接观察法和刃天青微孔板显色法。结果直接观察法和刃天青微孔板显色法判定所得的MIC结果具有高度的-致性。结论刃天青微孔板显色法具有结果客观、灵敏度高、准确等特点,因而可替代直接观察法用于真菌药敏实验终点的判定。
Objective To explore resazurin colorimetric microtiter plate assay in antifungal susceptibility testing for Candida spp. Methods Antifungal activity of itraconazol,fluconazole and amphotericin B against 87 clinical isolates of Candida spp were detected by reference method of NCCLS. The minimum inhibitory concentration (MIC)were determined by re- sazurin colorimetric microtiter plate assay and routine direct observing assay. Results Resazurin colorimetric microtiter plate assay and routine direct observing assay to determine endpoint were significantly identical. Conclusion Resazurin colorimetric microtiter plate assay is more objective and precise than routine direct observing assay;it can replace routine direct observing assay to determine antifungal endpoint.
