摘要
目的通过探讨串联质谱仪不同扫描方式对检测美国CDC酰基肉碱定值质控纸片结果的影响,寻找检测酰基内碱的最适扫描方式。方法分别采用母离子扫描法(precursorion,Prec)和多反应监测法(multiplereactionmonitoring,MRM)对美国CDC提供的酰基肉碱定值纸片(包括4个浓度水平)进行检测。每天测定所有水平的酰基内碱,连续测定5d。计算不同扫描方式测得的酰基肉碱的批内、批间CV%和相对偏倚。结果Prec法检测C2的批内CV%高于MRM法检测的批内C-W%(f=5.053,P=0.002);Prec法检测C6的批间CV%高于MRM法检测的批阃CV%0=2.737,P=0.034);Prec法和MRM法的相对偏倚均在美国CDC要求的范围之内。结论与Prec法比较,MRM法在检测个别酰基内碱时的精密度更好,但同时,优化MRM法也比较费时、繁琐。因此,建议将两种方法搭配使用,从而达到结果准确、省时省力、节约成本的综合效果。
Objective To find the best scan type of detecting acylcarnitines by analyzing the results of the quantitative dry blood spots of CDC in USA and exploring the influences. Methods All levels of acylcarnitines in the quantitative dry blood spots of CDC in USA were detected by precursor ion (Prec) and multiple reaction monitoring (MRM). There were four levels of the acylcarnitines. Detected all levels of acylcarnitines every day and there were five continuous days for de- tection. Calculated and analyzed the CV~ in the same day and in different days and bias of the results of acylcarnitines of different scan types. Results There were higher CV% in the same day of C2 detected by Pree than by MRM(t=5. 053,P=0. 002). There were higher CV% in the different days of C6 detected by Prec than by MRM (t = 2. 737,P= 0. 034). It could achieve the requirement of the CDC by the two scan types. Conclusion It is more precise for acylcarnitines to be de- tected by MRM than by Prec. But it need more time and more money for optimization of MRM. It is necessary to combine the two scan types for accurate results ,saving money and time.
