神经体液因子在兔血管迷走性晕厥中的变化

Variation of Neurohumor Factors in Rabbits with Vasovagal Syncope

摘要 目的通过兔急性失血建立血管迷走性晕厥(VVS)模型,研究其血浆肾上腺素(AD)、去甲肾上腺素(NE)、5-羟色胺(5-HT)、内皮素(ET-1)的变化,进一步了解VVS的发病机制。方法取幼年健康新西兰大白兔12只,通过股动脉放血建立VVS模型。使用50mL注射器以5mL/min速度抽血,连续监测其血压、心率,直至血压显著降低,神志丧失或抽搐。采用BL-420E+生物机能实验仪器监测,分别于静止后5min(T1),放血中升压最快点后1min(T2),降压最快点后1min(T3),呼吸、心跳消失或抽搐(T4)共取4次血样。比较各时间点兔血中AD、NE、5-HT、ET-1水平,并观察模型兔的最终状态。应用SPSS12.0软件进行统计学分析。结果在放血过程中各神经体液因子水平均呈上升趋势;在T2时,AD、NE、ET-1水平均明显升高(Pa<0.01);在T3、T4时5-HT水平明显升高(T3vsT1P<0.01,T4vsT3P<0.01)。实验结束时10只兔(83%)出现抽搐,最终血压、心率消失死亡。结论AD、NE、5-HT、ET-1等神经体液因子可能介导VVS的发病机制,为VVS的临床治疗和预防奠定了理论基础。 Objective To investigate the variation of neurohumor factors,including adnephrin(AD),norepinephrine(NE),5-hydroxytrypatamine(5-HT) and endothelin-1(EF-1) in order to understand the pathogenes of vasovagal syncope(VVS) in VVS mo-del of rabbits.Methods Twelve healthy New Zealand white rabbits were selected to establish VVS models.Fifty milliliter syringe was used to draw blood with the speed of 5 mL/min,continuous monitoring of blood pressure,heart rate,up to a significant reduction in blood pressure,loss of consciousness or convulsions. With BL-420E + experimental apparatus for monitoring biological functions,each rabbit received bloodletting at 4 time points:the point of 5 min after the static state(T1),the point of the fastest increasing pressure (T2),the point of the fastest decreasing pressure(T3),the point of the significant reduction in blood pressure,loss of consciousness or convulsions (T4).The concentrations of AD,NE,5-HT,ET-1 at T1,T2,T3,T4 were detected by ELISA and compared.The data were analyzed by SPSS 12.0 software.Results The concentration of neurohumor factors were all on the trend of rising during bloodletting.The concentration of AD,NE,ET-1 at T2 increased significantly (Pa〈0.01).The concentration of 5-HT at T3,T4 increased significantly (T3 vs T1 P〈0.01,T4 vs T3 P〈0.01).At the end of experiment,10 rabbits(83%) developed convulsion and eventually blood pressure and the heart rate vanished and died.Conclusions AD,NE,5-HT,ET-1 may play a part role in VVS.It provides a theory basis for the treatment and prevention of VVS.
