摘要
目的探讨多药耐药基因MDR1表达与耐药肝癌细胞株SMMC7721细胞周期的关系。方法血清饥饿法、药物双阻断法和分裂中期阻断摇落法同步化细胞;RT-PCR检测MDR1mRNA的表达。结果3种细胞同步化方法可分别得到高度同步化的G0/G1期、S期、G2/M期细胞;MDR1在SMMC7721细胞S期有表达,而在G0/G1和G2/M期无表达。结论S期MDR1基因的表达可能在肿瘤多药耐药出现的早期发挥了重要作用。
Objective To investigate the relationship between the expression of MDR1 gene and the cell cycle of hepatocellular carcinoma cells.Methods Three well established cell-cycle synchronization methods were used to synchronize tumor cells. RT-PCR was used to test the expression of MDR1 mRNA.Results Highly synchronized G0/G1,S,G2/M phase cells were obtainedby serum starvation, chemical block, or by combining mitotic shake-off with demeeolcine treatments. Conclusion The expression of MDR1 in S phase was involved in the early stage of multidrug resistance.
