摘要 目的:研究紫草复方的水提取工艺条件和高效液相色谱法同时测定咖啡酸四聚体、丹酚酸B含量的方法。方法:采用正交试验设计法,分别考察加水量、煎煮时间、煎煮次数对咖啡酸四聚体、丹酚酸B含量和浸膏得率的影响,分别赋予咖啡酸四聚体、丹酚酸B含量和浸膏得率不同的权重系数(依次为0.4、0.4、0.2),进行多指标综合评分。结果:测定的流动相为甲醇∶0.5%磷酸水=40∶60;流速1ml/min,检测波长252nm。优化工艺条件为加16倍量的水,煎煮3次,每次1h。结论:高效液相色谱法同时测定咖啡酸四聚体、丹酚酸B含量的方法准确,线性好,该工艺稳定、简便。 Objectives: To study the optimum condition for water extracting of complex Arnabia euchroma (Royle) Johnston and develop a HPLC method to simultaneously determine the contents of caffeic acid tettamer and salvianolic acid B. Methods: Orthogonal design was used, the effects of single parameters such as adding amount of water, time for boiling and boiling times, bestows on different weight coefficient for contents of caffeic acid tetramer and salvianolic acid B were 0.4, 0.4 and 0.2, were separately evaluated by synthesizing multiple guidelines method. Results: The mobile phase consisted of methol : 0. 5% phosphoric acid (40 : 60 v/v) at the flow rate of 1 ml/min, and the detecting wavelength was 252 nm. The optimum condition was as follows: the adding amount of water was 16 times of raw material, boiled 3 times, each time for 1 hour. Conclusions: The HPLC method for caffeic acid tetramer and salvianolic acid B is accuracy with better linearity, the technology for extracting is stable and convenient .
