黄芪总苷对兔动脉粥样硬化斑块中平滑肌表达的影响 被引量:4

摘要 目的:观察黄芪总苷对兔动脉粥样硬化斑块胶原含量的影响,并对其机制进行初步探讨。方法:高胆固醇饮食12周建立兔动脉粥样硬化(atherosclerosis,AS)病变模型,黄芪总苷10、30、90mg/kg·d灌胃。取主动脉内膜组织分别采用western、免疫组织化学方法检测斑块组织中Ras p21蛋白、SMCα-actin、c-jun、c-fos、c-myc蛋白含量。结果:黄芪总苷能明显降低斑块中SMCα-actin、Ras p21蛋白,c-jun、c-fos、c-myc蛋白和mRNA表达。结论:黄芪总苷能降低动脉粥样硬化斑块中平滑肌含量,其机制可能与降低Ras、c-jun、c-fos、c-myc表达有关。 Rabbit atherosclerotic models were set up to study effects of astragalosides on smooth muscle in atherosclerotic plaques and initial investigation for mechanism. Method: Rabbit atherosclerotic models were set up by hypercholestermia diet. Contents of Ras p21, SMC α-actin, c-jun, c-fos, c-myc protein were detected by western blot and immunohistochemical methods, respectively, contents of SMC α-actin, c-jun, c-fos, c-myc protein were measured by Image Pro Plus 4.5. Expression levels of c-jun, c-fos and c-myc mRNA were determined with real-time PCR. Result : In astragalosides groups, contents of Ras p21 and SMC α-actin protein, c-jun, c-fos, c-myc protein and mRNA were decreased markedly. Conclusion: astragalosides could decrease smooth muscle in rabbit atherosclerotic plaque. This maybe be concerned with that it could decrease expression of Ras, c-jun, c-fos and c-myc.
