褪黑素与氨基胍对内毒素血症山羊肝线粒体NO的影响

Effect of melatonin and aminoguanidin on nitric oxide in liver mitochondria of goats with endotoxemia

摘要 为探讨内毒素血症时山羊肝线粒体一氧化氮(NO)及一氧化氮合成酶(NOS)的变化,将72只山羊随机分成6组,每组12只,分别为A组对照组、B组内毒素血症模型组(内毒素,LPS 1 mg/kg体质量)、C组内毒素+褪黑素组(褪黑素,MT1 mg/kg体质量)、D组褪黑素组(MT1 mg/kg体质量)、E组内毒素+氨基胍组(氨基胍,AG25 mg/kg体质量)、F组氨基胍组(AG25 mg/kg体质量)。分别于处理后3 h和6 h各宰杀6只山羊提取肝线粒体,检测肝线粒体中NO、T-NOS和i NOS的变化。结果显示,山羊内毒素血症时肝脏线粒体NO水平升高,NOS活力增强。应用褪黑素和氨基胍治疗后,肝脏线粒体NO水平降低,NOS活力受到抑制,证明2种药物都具有一定的保护作用。 To investigate the effect of nitric oxide (NO) and nitric oxide synthase (NOS) on liver mitochondrial in gaots, 72 goats were divided randomly into 6 groups : A control, B lipopolysaccharide (LPS, 1 mg/kg BW), C lipopolysaccharide+ melatonin ( MT, 1 mg/kg BW), D melatonin ( MT, 1 mg/kg BW), E lipopolysaccharide + aminoguanidin (AG,25 mg/kg BW) and F aminoguanidin (AG,25 mg/kg BW). The liver mitochondrial NO,T-NOS and iNOS were examined at 3^rd h and 6^th after administration,respectively. Results revealed that NO and NOS increased in B group while decreased by the treatment of MT and AG. It is concluded that MT and AG could preserve the liver mitochondria by the inhibition of NO and NOS.
