摘要
目的探讨肝肿瘤细胞环氧合酶2(COX-2)和血管内皮生长因子(VEGF)关系。方法实验分为对照组和实验组,培养肝癌细胞株(BEL-7402)作为对照组,培养肝癌细胞株用不同浓度赛来昔布(celeccoxib商品名:西乐葆)处理作为实验组;提取mRNA,用ELISA方法检测细胞培养基中VEGF水平,用RT-PCR分别检测COX-2和VEGF的mRNA。结果正常培养的肝癌细胞能同时检测到COX-2和VEGF的mRNA;COX-2抑制剂赛来昔布能抑制VEGF的表达;COX-2在mRNA水平调控VEGF的表达。结论COX-2在VEGF的分泌与合成中起重要作用,从而影响肿瘤新生血管生成及肿瘤生长。
Objective To investigate the recationship between cvclooxygenase-2 (COX-2) and expression of vascular endothelial growth factor ( VEGF ) in hepatocellular carcinoma cell line BEL-7402. Methods mRNA was extracted from cultured hepatocellular carcinoma cell lines BEL-7402. The VEGF levels in the culture medium of these cells were measured using ELLSA. The expression of COX-2 and VEGF were identified by RT-PCR. Results The cultured hepatocellular carcinoma cell lines BEL-7402 can express both COX-2 and VEGF at same time. Celecoxib, COX-2 inhibitor, could decrease the expression of VEGF. And the levels of VEGF were correlated with COX-2 expression. COX-2 inhibited the expression of VEGF at mRNA level. Conclusions COX-2 plays an important role in the VEGF secretion and synthesis. Therefore, it has an effect on the angiogenesis and tumor growth.
