莲房原花青素对衰老小鼠脑组织中一氧化氮及一氧化氮合酶的影响 被引量:3

摘要 目的探讨莲房原花青素(lotus seedpod procyanidins,LSPC)对D-半乳糖衰老小鼠脑组织抗氧化系统的影响。方法60只昆明小鼠随机均分为5组:对照组(A组)、衰老模型组(B组)、LSPC低剂量组(C组,15mg/ml)、LSPC中剂量组(D组,30mg/ml)和LSPC高剂量组(E组,60mg/ml)。采用D-半乳糖建立小鼠衰老模型,化学比色法检测脑组织中总一氧化氮合酶(NOS)和神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)活性,一氧化氮(nitric oxide,NO)含量。用免疫组化法检测nNOS在脑组织中的表达。结果与B组相比较,C、D、E组LSPC能明显减少NO含量(P<0.05);D、E组LSPC不仅能明显降低总NOS和nNOS活性(P<0.01,P<0.05),且能减少nNOS在脑组织中的表达。结论LSPC对D-半乳糖所致脑衰老具有明显的保护作用。 Objective To study the effect of lotus seedpod procyanidins (LSPC) on anti-oxidative system in brain tissues of aging mice. Methods Sixty mice were randomly and evenly divided into five groups: negative control group (group A), aging group (group B), low LSPC group (group C, 15mg/ml), medium LSPC group (group D, 30mg/ml) and high LSPC group (group E, 60mg/ml). The senile model of mice was induced by D-gajactose. The content of nitric oxide (NO),activities of nitric oxide synthetase (NOS) and neuron nitric oxide synthetase (nNOS) of brain were measured by chemochromatometry methods. The expression of nNOS in brain was detected by immunohistochemical analysis. Results Compared with group B, NO contents decreased significantly in C, D and E groups (P〈0. 05). The activities of total NOS and neuron NOS of brain, and the expression of nNOS decreased significantly in D and E groups (P〈0. 01 or P〈0. 05). Conclusion LSPC has significantly protective effect of aging brain induced by D-gajactose.
