水红花子对小鼠免疫功能及迟发型超敏反应的抑制作用 被引量:7

摘要 目的研究水红花子对小鼠的免疫调节作用,并探讨其对迟发型超敏反应的影响.方法:用水红花子水煎剂对小鼠连续灌胃一周,采用MTT法检测脾淋巴细胞转化率、a-醋酸萘酯酶法检测T淋巴细胞数量,利用碳廓清实验检测吞噬细胞活性、定量溶血分光光度法检测IgM抗体生成量,并检测水红花子对DNCB诱导的迟发型超敏反应的影响.结果:与对照组相比,水红花子水煎剂可显著降低小鼠脾淋巴细胞的转化率,T淋巴细胞的数量、巨噬细胞的吞噬指数和IgM抗体生成量,可显著减轻耳部肿胀度,且差异均达到极显著水平.结论:水红花子可以明显抑制小鼠的细胞免疫和体液免疫功能,并能明显缓解由DNCB诱导的小鼠迟发型超敏反应. Objective.. The research is to study the immune regulate function of Fructus Polygoni Orentalis and explore its influence to the delayed type hypersensitivity(DTH) in mice. Methods.. The water decoction of Fructus Polygoni Orentalis is consecutively given to mice by means of intragastric perfusion for 7 days. Then MTT assay and acid naphthalene of a-acetic esterase of lymphocyte is used separately to test the lymphocyte transformation rate and the quantity of T cell. And the phagocytic function of macrophage is oberserved by carbon clearing test, the levels of IgM antibody are assessed through and the quantitative hemolysis spectrophotometrically. Then the effect of Fructus Polygoni Orentalis on DNCB-induced delayed type hyper- sensitivity is investigated. Results.- Compared with control group, Fructus Polygoni Orentalis decoction could markedly decrease the lymphocyte transformation rate, the quantity of T cell, phagocytic index of macrophage and the levels of IgM antibody. It also could markedly attenuate ear-swelling. Conclusion: Fructus Polygoni Orentalis can significantly inhibit cellular and humoral immune function and attenuate DNCB-induced delayed type hypersensitivity in mice.
