猕猴桃果王素对荷脂人脐静脉内皮细胞的影响

Effecs of kiwi Essence on lipid loaded human umbilical vein endotheliail cells

目的:观察猕猴桃果王素对ox-LDL诱导的荷脂人脐静脉内皮细胞(HUVECs)的影响。方法:将HUVEC与氧化低密度脂蛋白ox-LDL共孵育48h使其荷脂。然后分为四组进行处理,分别为对照组,10,20,40μmg/L猕猴桃果王素组。流式细胞仪检测细胞内脂质含量变化。RT-PCR检测ABCA1 mRNA表达,Western印记检测ABCA1蛋白表达。结果:猕猴桃果王素可明显降低荷脂HUVEC中脂质的含量,同时上调ABCA1的表达。两种作用均呈剂量依赖性。结论:猕猴桃果王素可明显减少ox-LDL诱导的HUVEC荷脂,这种作用可能与ABCA1的表达上调有关。

Objective： To observe the effecs of kiwi Essence on lipid loaded human umbilical vein endotheliail cells （HUVECs） induced by ox- LDL . Methods After incubated with ox-LDL for 48h, the HUVECs were divided into 4 groups as follows： control,10,20,40 μmg/L kiwi Essence.Cellular lipid content was determined by High Performance Liquid Chromatography （HPLC） .The mRNA level of ABCAI were determined by reverse transcription-polymerase chain reaction （RT-PCR） . The protein level of ABCA1 were determined by Western blot. Results kiwi Essence reduced the cellular lipid contents, and also upregulated the expression of ABCA1 with dose-dependent mannner. Conclusion kiwi Essence obviously decreased the lipid contents of HUVECs induced by ox-LDL,which may be related with the up-expression of ABCA1.
