凯时注射液对无心跳兔供体肺缺血再灌注损伤的影响 被引量:1

摘要 目的研究凯时注射液对无心跳兔肺缺血再灌注损伤的影响,并探讨其可能的作用机制。方法将24只健康新西兰大白兔随机平均分为三组,肝素化后处死,对照组立即予以LPD液灌注及保存,实验组1在处死后1h予LPD液灌注及保存,实验组2将凯时注射液(20ug/L)加入LPD液灌注及保存,两实验组在4℃保存5h后再灌注1h,对照组保存6h后灌注1h;测定经肺氧合后动脉血氧分压值(PaO2)和肺气道峰压值(PawP),于再灌注结束后取右上肺组织,测其湿重(W)与干重(D)、计算湿/干重比(W/D)、ELISA法检测超氧化物歧化酶(SOD)、丙二醛(MDA)含量,免疫组化法检测核因子-κB及ICAM-1的表达.并用光学显微镜和透射电子显微镜观察再灌注后肺组织结构的病理变化。结果再灌注45min后,实验组1的PaO2降低最明显、PawP升高最显著,实验组2及对照组的PaO2降低程度及PawP的升高程度均低于实验组1(P<0.01);实验组1的(SOD)明显低于对照组及实验组2,实验组1的(MDA)降低程度明显高于实验组2及对照组,实验组1的肺组织(W/D)、NF-κB及ICAM-1的表达高于对照组及实验组2(P<0.01),肺组织的病理变化实验组1最重。结论凯时注射液能减轻无心跳兔肺缺血/再灌注损伤,改善肺功能,具有肺保护功能。 Objective To investigate the protective effects of Lipo-PGE1 injection on non-heartbeat rabbit lung during isehemia-reperfusionl,and explore its possible meccthanism. Methods Twenty-four rabbits were randomly divided into three groups, after heparinization, The matched control immediately gives a LPD liquid to infuse to note and keeps, experiment set 1 the 1 h infuses to note and keeps to the LPD liquid after sentencing to death, experiment set 2 Lipo-PGE1 (20 ug/L) to join a LPD liquid to infuse to note and keep, two keep a 5 hs in 4 ℃ after infuse again to note a 1 h, the matched control infuses to note after keeping the 6 hs 1 h ; After lung oxygen matches measurement artery blood oxygen cent presses a value( PaO2 ) the way the Feng presses a value(PawP) with lung spirit, in again infuse and note be over behind take right top lung organization, measure it wet heavy(W) and dry heavy(D), compute wet/dry heavy ratio( W/D), ELISA method's examining super oxide Qi and turning Mao(SOD) and C is two Quans(MDA) content, immunity set the expression that turn the method examination pit faetor-κB and ICAM-1. Counteract an optical microscope and deeply shoot an electronic microscope observation again infuse to note behind lung organization structure of pathologic variety. Results After infusing again to note a 45 mins, experiment set one PaO2 lowers most obviously, PawP goes up to most , experiment set 2 and the PaO2 of matched control's lowering degree and going up of PawP degree is aller low than an experiment set 1 ; (P 〈0. 01 ) Experiment set 1 of(SOD) is more obviously low than the matched control and the experiment set 2, experiment set 1 of(MDA) lower degree is more obviously high than an experiment set 2 and matched control, the expression of experiment set one lung organization (W/D), the NFηκB and ICAMIj1 is higher than the matched control and the experiment set 2 (P 〈 0. 01 ), lung organization of pathology variety experiment set 1 the most heavy. Conclusion Lipo-PGE1 injection can alleviate the ischemia effect of non-heartbeat rabbit lung preservation.
