摘要
目的探讨用中药水蛭、银杏、川芎的有效成分组成的复方制剂对大鼠缺血再灌注损伤脑组织内肿瘤坏死因子-a(TNF-a)表达的影响。方法将60只大鼠随机分为A、B两组,制作脑缺血再灌注动物模型。A组给予复方制剂,B组给予生理盐水。然后应用ELISA法,测定不同时间点时两组大鼠脑梗死灶内TNF-a的含量。结果缺血前,两组大鼠脑组织内TNF-a均呈低水平表达。两组相比,差异不明显(P>0.()5)。缺血3 h再灌注3 h、6 h、24 h、72 h时,两组大鼠缺血侧脑组织内的TNF-a均呈高水平表达,但A组鼠脑组织内的TNF-a表达低于B组的,差异具有显著性(P<0.05)。结论用中药水蛭、银杏、川芎的有效成分组成的复方制剂不影响正常脑组织内低水平的TNF-a的表达,但能抑制缺血再灌注损伤脑组织内高水平的TNF-a的表达,对脑组织具有一定的保护作用。
Objective To observe the influence of compound agent made up with effective ingredients of Hirudo and Rhizoma ehuanxiong and Folium Ginkgo on the expression of tumor necrosis factor( TNF - a) in rats cerebral tissue of undergoing ischemia reperfusion. Methods We randomly decided the 60 rats into A group and B group, made the models of cerebral isehemia reperfusion, administered compound agent to A group of rats and normal saline to B group of rats, then, at various time points, measured the quantifies of TNF - a in cerebral tissue of the two groups of rats by using ELISA method. Results Before cerebral ischemia, the expression of TNF - a in cerebral tissue of the two groups of rats was low level, the difference had no significance when comparing between A geoup and B group( P 〉 0. 05 ). At reperfusion for 3 h ,6 h ,24 h and 72 h fallowing isehemia for 3 b, the expression of TNF - a in the brain tissue of the two groups of rats was high level, but the exp ion of TNF - a in A group was dropped when comparing with that of B group( P 〈 0.05 ). Conclusion The compound agent made up with effective ingredients of Hirodo and Rhizoma chuanxiong and Folium Ginkgo agent does not influence the low - level expression of TNF -a in normal brain tissue, but can significantly restrain the high -level epression of TNF -a in cerebral tissue of ischemia - reperfusion in - injury, it has protective effect on the brain tissue.
