摘要
目的:观察香烟烟雾提取物(cigarette smoke extract,CSE)对人气道平滑肌细胞(human airway smooth muscle cells,HASMCs)的损伤及对白介素-8(interleukin-8,IL-8)表达水平的影响。方法:原代培养HASMCs,分为空白对照组、CSE组,在干预24h后使用倒置显微镜、MTT法观察HASMCs的活性,用ELISA法和RT-PCR法测定IL-8表达水平的变化。结果:CSE可导致HASMCs形态和结构的改变,同时使HASMCs分泌释放IL-8增加。结论:在香烟烟雾提取物刺激下,HASMCs分泌IL-8增加。
Objective To investigate the effects of cigarette smoke extract (CSE) on human airway smooth muscle cells (HASMC) and on the expression of interleukin-8 (IL-8) in them. Method The primary cultured HASMC were divided into blank control group and CSE group. Then the activity of HASMC was observed by invert microscope and MTT method, and the expression of IL-8 was measured by ELISA and RT-PCR 24 h after intervention. Result CSE can cause morphologic and structural changes of HASMC and increase the expression of IL-8 in HASMC. Conclusion Under the stimulation of CSE, expression of IL-8 is increased in HASMC.
