高效液相色谱法测定双黄连栓中金银花的绿原酸含量

摘要 [目的]建立双黄连栓中绿原酸含量的HPLC测定方法。[方法]色谱柱为Diamonsil C185μm(250 mm×4.6 mm)色谱柱,流动相为甲醇-水-冰醋酸-三乙胺(15.0∶85.0∶1.0∶0.3),检测波长为324 nm,流速为1.2 ml/min,进样量为20μl,柱温25℃,在该条件下测定双黄连栓中金银花的绿原酸含量。[结果]试验条件下,绿原酸在0.2~0.6μg范围内线性关系良好,r=0.998 7;样品在5 h内稳定;平均回收率99.6%,RSD=0.8%(n=6)。[结论]该方法简便、快速、准确,可用于双黄连栓的含量测定及质量控制。 [ Objective ]To establish a method for the determination of Chlorogenic acid content in Shuanghuanglian suppository. [ Method] The chromatographic column was Diamonsil C18 5μm(250 mm ×4.6 mm), The mobile phase was methanol - water - glacial acetic acid - triethyl- amine (15.0:85.0:1.0:0.3) , the detection wave length at 324 nm and the flow rate was 1.2 ml/min. [ Result ] Chlorogenic acid had a good relationship in the range of 0.2 ×0.6 μg, r = 0.998 7 . The average recovery was 99.6% and RSD = 0.8% ( n = 6). [ Conclusion ] This method is easy, sensitive and accurate for the content determination of Shuanghuanghan suppository and quality control.
