趋化因子受体P2Y与鼻咽癌转移相关性初步研究 被引量:1

摘要 目的探讨P2Y受体在鼻咽癌细胞中的表达及其与鼻咽癌转移之间的关系。方法采用RT-PCR检测P2Y受体在鼻咽癌细胞系中的表达,趋化试验分析P2Y受体与鼻咽转移的关系。结果P2Y受体在7株鼻咽癌细胞中的表达有明显的差异,其中P2Y12在所有鼻咽癌细胞株中均无表达,而P2Y1和P2Y6仅弱表达于HNE2细胞中,在其他细胞株中不表达。HNE2细胞株几乎表达所有检测的P2Y受体,而HNE3细胞株仅表达P2Y11受体。趋化试验表明,鼻咽癌细胞能在ATP作用下发生明显移动,以ATP浓度为1.0×10-6mol/L时趋化作用最明显,其趋化指数(CI)为4.3±0.6,这种趋化作用是一种定向迁移,而非随机运动。结论P2Y受体在鼻咽癌的转移中可能发挥着重要的作用。 [Objective] To explore the expression of P2Y receptors in nasopharyngeal carcinoma cells in order to reveal the relationship between the P2Y receptor and the migration of nasopharyngeal carcinoma. [Method] The expression of P2Y receptors in 7 nasopharyngeal carcinoma cells were detected by RT-PCR. And the migration of nasopharyngeal carcinoma cell CNE2 induced by ATP were analysed. [Results] There were significant difference of the P2Y receptors'expression in the nasopharyngeal carcinoma cells. P2Y12 did not expressed in all nasopharyngeal carcinoma cell lines. P2Y1 and P2Y6 only weakly expressed in HNE2 cell hne, no expression in other cell lines. Almost P2Y receptors expressed in HNE2 cell line, and only P2Y11 expressed in HNE3 cell line. Chemotaxis test showed that nasopharyngeal carcinoma cells could migrate induced by ATP, the chemotactic role was most obvious at 1.0×10^-6 mol/L of the concentration of ATP. Its chemotactic index (CI) was 4.3±0.6, which is a migration of chemotaxis, instead of random movement.[Conclusion] The expression of P2Y receptors possibly played a vital role in the metastasis of nasopharyngeal carcinoma.
