摘要
为探索打顶促进烟株体内烟碱合成的调控机制以及茉莉酸(JA)信号途径在其中的作用,用JA处理模拟烟草打顶,采用差异蛋白质组学技术对烤烟根尖组织细胞蛋白质组进行分析.2-DE图谱经ImageMaster 2DPlatinum(Version 5.01)软件识别结果显示,约有500个蛋白质点被清楚分离,通过比较发现,打顶和JA处理后表达上调的蛋白质分别有13和26个,其中有3个共同蛋白质点;表达下调的分别有5和6个,其中有3个共同蛋白质点;特异表达点分别有8和5个,其中有4个共同蛋白质点.将其中8个差异点采用基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)进行了肽质量指纹图谱分析,Mascot软件对NCBInr数据库查询,8个特异蛋白质点得到了鉴定.其中4个蛋白质点在JA处理和打顶处理中的变化是相同的,表明这些蛋白质点可能受到茉莉酸信号途径的调控,同时又是刺激烟碱合成的重要因子.
To study the molecular mechanism of nicotine synthesis stimulated by topping and the action of JA Signaling pathways on it. We simulated flue-cured tobacco topping with JA. The proteomic of flue-cured tobacco root tip were analyzed by technology and method of differential proteomics. The resuits analyzed by Image Master 2 D software showed that there were about 500 detectable spots which were separated distinctly. By comparison, the up proteins in topping and JA treatement were thirteen and twenty-six, and there were three common spots. The down proteins were five and six, and there were three common spots. Eight and five differential expressed spots were found respectively, and there were four common spots. With direct MALDI - TOF mass spectrometry analysis and protein database searching, eight protein spots were identified, in which four differential proteins showed the same trend in topping and JA treatment, suggesting that they act as nicotin-regulated biosynthesis.
