NRDS患儿外周血单核细胞NF-κB活性及血浆TNF-α变化及其意义 被引量:1

Changes and clinical significance of the peripheral blood mononuclear cell NF-κB activity and TNF-α in neonatal respiratory distress syndrome

摘要 【目的】探讨新生儿呼吸窘迫综合征(neonatal respiratory distress syndrome,NRDS)患儿外周血单核细胞核因子-κB(NF-κB)及血浆肿瘤坏死因子-α(TNF-α)的表达及其临床意义。【方法】分别采用免疫组织化学染色法和酶联免疫吸附法测定54例NRDS患儿及35例正常对照组的外周血单核细胞NF-κB活性和血浆TNF-α水平,并分析NF-κB活性与TNF-α浓度相关性。【结果】NRDS组NF-κB活性和TNF-α水平显著高于正常对照组(P<0.01),而且NRDS外周血单核细胞NF-κB活性与TNF-α水平显著正相关。【结论】新生儿呼吸窘迫综合征外周血单核细胞核因子-κB活性明显升高,NF-κB是NRDS患儿不稳定的标志,并可能通过上调TNF-α水平而促发NRDS。 [Objective] To investigate the clinical significance of changes of the activity of peripheral blood mononuclear cell NF-kB and TNF-α in neonatal respiratory distress syndrome(NRDS). [Methods] Enzyme linked immunosorbent assay was used to measure the plasma TNF-α and immunohistochemical method was used to measure activity of NF-kB in peripheral blood mononu-clear cell in 54 patients with NRDS and 35 normal control subjects. At the same time linear correlation analysis was used to analyze the correlation between TNF-α and NF-kB. [Results] The level of activity of NF-kB in peripheral blood rnononuclear cells and the plasma TNF-α cocentration were significantly higher in patients with NRDS than those in the controls. The level of plasma TNF-α was positively correlated with activity of NF-kB in peripheral blood mononuclear cells. [Conclusions] The activity of NF-kB in peripheral blood mononuclear cells is significantly elevated in NRDS. It is a potential marker of plaque unstahility and may induce NRDS via up-regulating TNF-α expression.
