长双歧杆菌BBMN68工业化培养基筛选 被引量:6

摘要 本实验以长双歧杆菌BBMN68生长状况为评价指标筛选一种低成本、易配制的工业化培养基。实验比较了菌株在四种常用双歧杆菌培养基中的生长状况,结果表明NPNL培养基生长效果最好(P<0.05);通过低成本氮源代替NPNL氮源,发现玉米浆干粉培养效果最好(P<0.05),玉米浆干粉最佳用量为55g/L;玉米浆干粉培养基中剔除磷酸盐缓冲液、无机盐溶液、L-半胱氨酸盐酸盐,对BBMN68生长状况没有显著性影响(P>0.05)。长双歧杆菌BBMN68工业化最佳培养基配方为:55g/L玉米浆干粉,10g/L葡萄糖,1g/L吐温-80,活菌数达到4.4×109cfu/mL。 This study was aimed to screen an industrialized culture media which could be low-cost and easy to prepare for the growth of Bifidobacterium longum BBMN68. Comparing with other bifidobaeterial culture medium, NPNL was proved to be the best Culture Media for BBMN68. It was shown that corn steep liquor dry meal was the best replaced nitrogen source compared with skim milk powder and lactalbumin, and the best concentration was 55g/L. The viable counts of BBMN68 was not primarily affected with the absence of the phosphate buffer solution, inorganic salt solution and L-cysteine hydroehloride. The optimized composition was determined as follows: corn steep liquor dry meal 55g/L, glucose 10g/L and tween-80 1g/L. The total viable counts can increase to 4.4×10^9cfu/mL.
