摘要
目的探讨结核分枝杆菌CFP10-ESAT6融合蛋白对不同分枝杆菌致敏动物的鉴别作用。方法以不同分枝杆菌分别致敏豚鼠,致敏成功后,每只豚鼠皮内分别注射结核分枝杆菌CFP10-ESAT6融合蛋白和结核菌素纯蛋白衍生物(PPD)。用双盲法记录注射后24和48h注射局部硬结的纵径和横径,计算其均值。结果PPD对结核分枝杆菌活菌、死菌和卡介苗致敏豚鼠的皮肤反应均为阳性,局部硬结直径分别为(13.91±1.04)mm、(13.11±1.40)mm和(13.16±1.43)mm。结核分枝杆菌CFP10-ESAT6融合蛋白对结核分枝杆菌活菌致敏豚鼠的皮肤反应为强阳性(≥10mm),局部硬结直径为(10.53±2.66)mm;对结核分枝杆菌死菌致敏豚鼠的皮肤反应为弱阳性(≤5mm)或阴性,局部硬结直径为(1.61±1.39)mm;对卡介苗致敏豚鼠的皮肤反应为阴性,局部硬结直径为0。结论结核分枝杆菌CFP10-ESAT6融合蛋白可鉴别结核分枝杆菌活菌、死菌和卡介苗致敏动物。
Objective To investigate the differentiation of animals sensitized with various Mycobacterium tuberculosis by M. tuberculosis CFP10-ESAT6 fusion protein. Methods Guinea pigs were sensitized with various M. tuberculosis, then injected i.d. with CFPIO-ESAT6 protein and PPD. The longitudinal and transverse diameters of local indurations at injection sites were measured by double-blind method 24 and 48 h after injection, and the mean diameter was calculated. Results All the skin test results of PPD in guinea pigs sensitized with live and inactivated M. tuberculosis and BCG were positive, and the mean diameters of local durations were (13.91 ±1.04), (13. 11 ±1.40) and (13. 16 ±1.43) mm respectively. However, CFP10-ESAT6 protein was strong positive in skin test in guinea pigs sensitized with live M. tuberculosis, weak positive or negative in those with inactivated M. tuberculosis, and negative in those with BCG, and the corresponding mean diameters of local durations were ( 10. 53 ±2.66), ( 1.61 ±1.39 ) and 0 mm respectively. Conclusion M. tuberculosis CFP10-ESAT6 protein may be used for the differentiation of animals sensitized with live and inactivated M. tuberculosis and BCG.
