摘要
目的:观察叶下珠复方Ⅱ号抗大鼠实验性肝纤维化的作用效果并探讨其作用机制。方法:采用二甲基亚硝胺诱导的大鼠肝纤维化模型,用高、中、低三种剂量(100g/kg,50g/kg,25g/kg)的叶下珠复方Ⅱ号水提液进行干预治疗,并与秋水仙碱进行对照,检测大鼠肝功能、肝组织羟脯胺酸(Hyp)、丙二醛(MDA)、血清透明质酸(HA)含量及肝脏病理组织学改变。结果:叶下珠复方Ⅱ号高、中、低剂量组谷草转氨酶(AST)、γ-谷氨酰转移酶(GGT)、碱性磷酸酶(ALP)、总胆红素(TBIL)和直接胆红素(DBIL)均显著低于模型组,差异均有显著性意义(P<0.05)。叶下珠复方Ⅱ号中剂量组大鼠肝组织Hyp、MDA和血清HA含量均明显低于模型组,差异均有显著性意义(P<0.05);与秋水仙碱组比较,差异均有显著性意义(P<0.01)。叶下珠复方Ⅱ号高、中、低剂量组肝组织纤维化病变明显减轻,与模型组比较,差异均有显著性意义(P<0.01)。结论:叶下珠复方Ⅱ号对二甲基亚硝胺诱导的肝纤维化形成具有明显的抑制作用,其机制与抗脂质过氧化损伤、保护肝细胞和抑制胶原纤维合成有关。
Objective:To investigate the prophylactic effect of Compound Phyllanthus Urinaria Ⅱ( CPU Ⅱ) on experimental liver fibrosis in rats induced by dimethylnitrosamine(DMN) and to reveal its mechanism. Methods: By establishing the models of DMN-induced liver fibrosis in rats, we observed the prophylactic effect of CPU II with high, middle and low dosage group( 100g/ kg,50g/kg and 25g/kg respectively. ), and colchicinum as the positive control. Serum Index of liver function, contents of hydroxyproline(Hyp) ,malodialdehyde (MDA) in liver tissue and serum hyaluronic acid(HA) were determined,the liver pathological change was observed. Resu/ts :It showed that serum levels of AST,ALP,GGT,TBIL and DBIL in high, middle and low dosage groups of CPU Ⅱ were all obviously lower to those of model group(P 〈0. 05). Contents of Hyp,MDA in liver tissue and HA in serum were remarkable lower to those of model group ( P 〈 0. 05 ) and colchicinum group ( P 〈 0. 01 ). Hepatic fibrosis histopathological changes were remarkable alleviated in three dosage groups of CPU Ⅱ comparing with those of model group ( P 〈 0. 05 ). Conclusion: It showed that CPU Ⅱ has a remarkable prophylactic effect on DMN-induced liver fibrosis in rats, probably by protecting hepatocyte and suppressing fibrosis accumulation through anti-lipopeioxidation.
