重组丝氨酸羟甲基转移酶活性测定及酶促反应条件的优化研究

The Activity Assay of Recombined SHMT and the Optimization of the Enzymatic Reaction

本文采用了高效液相色谱法测定重组丝氨酸羟甲基转移酶活力 ,并利用该法将一系列的反应条件 (温度、p H、辅因子浓度等 )加以优化 ,从而确定了该酶促反应的最佳反应条件。在该反应条件下 ,对底物甘氨酸的转化率可达 78%。

The analytical method of determing recombined SHMT activity with HPLC was founded and a series reaction conditions such as tempreture, pH and cofacter concetration etc. were optimized. Thus the optimal enzymytic reaction condition was obtained. Under this condition, the transformation rate can approach 78%.
