骨桥蛋白在骨愈合过程中的基因表达 被引量:8

摘要 目的:研究骨桥蛋白mRNA在骨愈合过程中表达的变化。方法:24只雄性SD大鼠胫骨上端造成不全骨缺损做为动物模型,采用原位杂交技术观察骨桥蛋白mRNA在骨愈合过程中不同时期基因表达情况。结果:术后1周时在近髓腔侧开始出现编织骨,骨桥蛋白mRNA在成骨细胞和骨细胞中呈强表达。术后第3周时,缺损区被骨痂填满且板层骨增多,编织骨明显减少,表达骨桥蛋白mRNA的细胞数也相应减少。结论:骨桥蛋白主要表达在骨痂形成早期。 ABSTRACT Objective: To observe changes of the mRNA of osteopontin ( OP ) during the bone healing process. Methods: 24 male SD rats were used and the experimental model was made by digging a defect in the upper tibia. The expression of OP mRNA was studied by in situ hybridization during the various stages of bone repairment. Results: Newly formed woven bone began to appear near the medullary cavity 1 week after the operation, the mRNA was highly expressed in the osteoblasts and osteocytes. 3 weeks later, the defects were fully filled with callus, in which the lamellae bone increased and the woven bone declined in the meantime. The cells expressing the OP mRNA declined also. Conclusion: The OP were mainly produced in early phase of bone callus formation when the osteoblasts could transcript it intensively.
