正常胎盘及葡萄胎滋养细胞H-ras和C-myc基因产物表达 被引量:3

H-ras and C-myc gene expression in normal human placenta and in hydatidiform mole trophoblast.

摘要 目的:探索ras、myc基因蛋白在正常胎盘发育中的作用及ras、myc基因蛋白表达与葡萄胎临床预后关系。方法:用免疫组化方法,分析40例正常胎盘及50例葡萄胎组织H-ras和C-myc基因蛋白表达情况。结果:孕早期胎盘ras、myc基因蛋白的表达部位主要在细胞滋养细胞,表达率分别为70%和80%,孕中期表达部位主要在合体滋养细胞,表达率逐渐降低,足月时表达消失。葡萄胎ras、myc基因蛋白表达率分别为52%和60%,myc基因蛋白表达与滋养细胞的增生程度有关(P<001),ras、myc基因蛋白表达与葡萄胎恶变有关(分别P<001,P<005)。结论:ras、myc基因蛋白的过度表达与葡萄胎恶变有关。 Objective:To explore the effect of ras,myc gene proteins on the development of placenta and the value of ras,myc expression in clinical prognosis of hydatidiform mole.Methods:Immunocytochemistry was used to locate H-ras,C-myc protein in 40 normal placental specimens and 50 hydatidiform mole samples.[WTHZ]Results:The rates of ras and myc gene protein expression in first trimester placenta were 70% and 80% respectively.The immunocytochemical localization of ras,myc protein was mainly in cytotrophoblast.The immunostaining of ras and myc steadily decreased in second trimester.The immunocytochemical localization was mainly in syntrophoblast.Immunoreactivity for ras,myc protein disappeared term.The rate of ras and myc protein expression in hydatidiform mole was 60% and 62% respectively.There was a strong association between trophoblastic proliferative grade and myc expression(P<0.01).The expression of ras,myc protein associated with the malignance of hydytidiform mole(P<0.01,P<0.05 respectively).Conclusion:Ras and myc gene proteins play the role in the control of both the proliferative and differentiation pathways of trophoblastic cells of normal placenta and in the malignant mechanism of hydatidiform mole.The overexpression of ras and myc proteins associated with the prognosis of hydatidiform mole.
