重组痘苗病毒介导的IL-2抗小鼠FBL-3细胞肿瘤的实验研究

摘要 观察了编码人白细胞介素2(IL-2)基因的重组痘苗病毒(recombinantvacciniavirusencodinghu-manIL-2gene,IL-2VV)对小鼠接种FBL-3红白血病细胞诱发肿瘤的治疗作用。野生型FBL-3红白血病细胞接种C57BL/6小鼠皮下,建立荷瘤模型,随机分组,分别局部注射IL-2VV、TKVV、重组IL-2、Hank's液0.2ml/次,3/周,连续治疗2周。结果发现,小鼠局部注射IL-2VV组肿瘤出现晚,生长速度缓慢,生存期延长,与对照组相比,差异有非常显著性意义。进一步观察还发现IL-2VV能显著提高荷瘤小鼠脾细胞的自然杀伤细胞(NK)、细胞毒T淋巴细胞(CTL)的活性,明显增强腹腔巨噬细胞杀伤活性。 The therapeutic effect of recombinant vaccinia virus encoding human IL 2 gene(IL 2VV) on the tumor bearing mice was investigated. The tumor bearing mice were established by subcutaneus inoculation with 2×106 FBL 3 erythroblastic leukemia cells into C57BL/6 mice. The mice were injected in situs with IL 2VV, TKVV, IL 2 and Hank's solution three times a week for 2 weeks, respectively. The results showed that IL 2VV treatment could significantly inhibit the tumor growth and prolong the survival duration of the tumor bearing mice. Further study demonstrated that the NK, CTL activities and macrophage cototoxicity were significantly augmented after treatment with IL 2VV. These results suggested that IL 2VV could effectively increase the anti leukemia immunological response and might be of great importance for the study of gene therapy of leukemia.
