胃癌组织bcl-2蛋白表达和DCC基因的杂合性缺失 被引量:1

摘要 为了研究bcl2蛋白表达和DCC基因的杂合性缺失(LOH)在胃癌发生中的作用,采用免疫组织化学技术和PCR方法检测了胃癌组织bcl2蛋白的表达和DCC基因的杂合性丢失(LOH)。结果发现,胃癌(信息个体)LOH发生率为54.5%(18/33),bcl2蛋白的表达阳性率为60.0%(30/50),Ⅲ、Ⅳ期胃癌LOH发生率(83.3%,15/18)显著高于Ⅰ、Ⅱ期胃癌(20.0%,3/15),P<0.01。bcl2蛋白的表达和DCC基因的LOH与胃癌大小、分化程度、淋巴结转移、浆膜浸润、临床分期及Lauren′s分型无显著相关。结果提示bcl2蛋白的高表达参与了胃癌的发生,DCC基因的LOH与胃癌的进展有关。 In order to investigate the roles of expression of bcl-2 protein and LOH at DCC gene in the carcinogenesis of gastric cancer,the expression of bcl-2 protein and LOH at DCC gene were detected by immunohistochemical technique and PCR method.LOH was detected in 18/33(54.5%) of gastric cancers,bcl-2 protein was detected in 30/50 (60.0%).The frequence of LOH in stage Ⅲ~Ⅳ gastric cancer was significantly higher than that in Ⅰ~Ⅱ stage (P<0.01).The expression of bcl2 protein was not associated with LOH at DCC gene,tumor size,differentiation,lymphnode metastasis,serosal invasion,clinical stage and Lauren′s type.The results suggest that high expression of bcl-2 protein is involved in the development of gastric cancer and LOH at DCC gene is involved in its progression.Their carcinogenetic mechanism milght be different.
