摘要
为了解胃癌组织中多种基因蛋白异常表达及意义,应用免疫组化技术检测51例手术切除胃癌组织标本中增殖细胞核抗原(PCNA)及P53、K-ras、Bcl-2蛋白的表达。结果胃癌组织中PCNA标记指数(PCNALI)为51.2±20.5,P53,K-ras,Bcl-2蛋白表达率分别37.3%,17.6%和68.6%,均显著高于正常胃粘膜(P<0.05)。PCNALI与肿瘤大小、淋巴结转移及临床分期密切相关(P<0.05)。肠型胃癌中K-ras、Bcl-2蛋白表达率分别为43.8%和93.8%,均显著高于胃型胃癌的6.0%(P<0.01),54.5%(P<0.05),而PCNALI及P53蛋白表达率两型胃癌无显著差别。提示PCNA、P53、K-ras、Bcl-2蛋白异常表达与胃癌的发生发展有关。PCNALI可能对判断胃癌的预后有一定价值。K-ras。
In order to investigate the role of multiple genetic expression in the development and progress of gastric carcinoma(GC), the expressions of PCNA, P53, K ras, Bcl 2 proteins were detected in 51 surgically resected GC specimens with immunohistochemical technique. It was found that PCNALI in GC was 51 2±20 5 and expressions of P53, K ras, Bcl 2 proteins were 37 3%, 17 6% and 68 6%, respectively. PCNALI was positively correlated with tumor size, lymphonode metastesis and clinical stages( P <0 05). The expression rates of K ras(43 8%) and Bcl 2(93 8%)in intestinal type GC were significantly higher than those of diffuse type of GC(6 0%, 54 5%) ( P <0 01, P <0 05), however no significant difference of P53 protein expression and PCNALI was found between the two type GC. These findings suggested that the abnormal expressions of PCNA, P53,K ras,Bcl 2 were corelated with the development and progress of GC and PCNALI may be of some value for predicting prognosis of GC. K ras, Bcl 2 may be two susceptable target genes of GC.
