摘要
目的验证K-ras基因突变与化疗敏感性之间关系。方法用MTT法测定了17种抗癌药对43例大肠癌的抑制率,并用聚合酶链反应—限制性片段长度多态性分析法检测大肠癌中K-ras基因突变情况,将其分成K-ras突变组与正常组,分别统计17种抗癌药在两组中抑制率大小,并进行比较。结果除丝裂霉素、平阳霉素、顺铂外,抗癌药在正常组中的抑制率大于在突变组中的抑制率,其中更生霉素,甲氨喋呤、长春新碱、鬼臼乙叉苷、阿霉素、阿克拉霉素两组存在显著差异。结论肠癌中K-ras基因存在突变时,抗癌药耐药性增加。
おURPOSE To testify the relationship between K-ras gene mutations and chemosensitivity. METHODS We tested the inhibition rates of 17 antitumor drugs in 43 colorectal cancer patients using MTT method, and also analysed the condition of K-ras gene mutation with polymerase chain reaction-restriction fragment length poymorphism in it. We divided these patients into K-ras gene mutational group and normal group,and calculated the inhibition rates of the antitumor drugs in the two groups,compared the results.RESULTS It demonstrated that except MMC,BLEO,DDP,the inhibition rates of the antitumor drugs in normal group were higher than in mutational group,and the significant difference existed between the two groups in DTC,MTX, VCR, VP-16,DXR,ACM.CONCLUSIONS When K-ras gene mutations are present in the colorectal cancers, the antitumor drugs' resistence occur.
