摘要
应用原位分子杂交、免疫组化、免疫荧光定量分析的方法研究CD44V6在人大肠癌细胞中的表达,应用激光漂白后荧光恢复技术研究CD44V6对大肠癌细胞的缝隙连接细胞间通讯的影响。结果表明,CD44V6在HT29、Lovo细胞中均有表达,不管在mRNA水平还是在蛋白质水平,高转移能力的Lovo细胞表达CD44V6均高于低转移能力的HT29细胞。将HT29、Lovo细胞用CD44V6单抗封闭则发现CD44V6不能使HT29、Lovo细胞的缝隙连接细胞间通讯发生明显改变。说明CD44V46的表达与大肠癌细胞的转移潜能有关。
Expression of CD44V6 variant in human colorectal carcinoma HT29 and Lovo cell lines was investigated by in situ hybridization, immunohistochemistry and quantitative immunofluorescenece technique at mRNA and protein level. The effect of CD44V6 variants on gap junctional intercellular communication (GJIC) was studied by fluorescence recovery after photobleaching (FRAP). The result showed that both of HT29 and Lovo cell lines were well expressed in the CD44V6 mRNA and their proteins, but Lovo cells expressed more highly than HT29 cells both at mRNA and protein levels. Moreover, the expressions of CD44V6 were different among Lovo cell lines, part of which expressed more.The expression of CD44V6 variant was related to metastatic potential of human colorectal carcinoma cells,but CD44V6 could not affect the GJIC of HCC cells.
