摘要
目的:探讨清气化痰汤对慢性支气管炎(慢支)大鼠气道黏液高分泌模型的干预作用。方法:24只Wistar大鼠随机分为3组(n=8):空白对照组、模型组、清气化痰汤组。经气道注入脂多糖及烟雾吸入复制慢支大鼠气道黏液高分泌模型。采用免疫组化法检测肺内气道的Muc5ac蛋白表达,RT-PCR法检测肺组织Muc5acmRNA;ELISA法检测支气管肺泡灌洗液中的TNF-α和IL-8浓度。结果:(1)模型组大鼠肺组织中Muc5acmRNA表达明显高于空白对照组;与模型组比较,清气化痰汤组显著降低(P<0.05),肺内气道上皮Muc5ac水平的改变与Muc5acmRNA改变相似。(2)模型组TNF-α和IL-8增加,清气化痰汤干预后TNF-α和IL-8均明显降低。(3)BALF中TNF-α浓度与Muc5acmRNA的表达呈正相关;IL-8浓度与黏蛋白Muc5acmRNA的表达呈正相关。结论:清气化痰汤明显抑制慢支大鼠气道黏液高分泌状态,其机制可能与TNF-α或IL-8被抑制有关。
Objective:To investigate the effects of Qingqihuatan Tang in airway mucus hypersecretion in rat model of chronic bronchitis (CB). Methods :24 Wisatr rats were randomly divided into 3 groups (n = 8 in each group), Qingqihuatan Tang intervention group,model group( LPS-cigarette smoking induction group)and control group. MucSac and its mRNA expression were assayed by immunohistochemistry and RT- PCR, ELISA technique was performed to detect TNF-α and IL -8 in bronchoalveolar lavage fluid(BALF). Results:LPS-cigarette smoking induced the expressions of Muc5 ac mRNA. And protein in bronchial epithelia, and increased the release of TNF - α and IL - 8 in BALF. The up-regulated expression of MucSac mRNA Was positively correlated with IL - 8 and level of TNF -α ( both P 〈 0.05 ). Qingqihuatan Tang significantly attenuated bronchial MucSac expression and reduced the level of TNF - α and IL - 8 . Conclusion: The inhibitory effect of Qingqihuatan Tang may be attributed to the reduction of IL - 8 andTNF - α released in the lung.
