两种蝮蛇毒的蛋白质组凝胶内差异显示电泳分析

摘要 目的:建立具有高分辨率和稳定性的江浙蝮蛇毒与白眉蝮蛇毒蛋白质组的双向电泳图谱,并对其进行差异蛋白质组分析。方法:取江浙蝮蛇毒与白眉蝮蛇毒,分别用Cy3和Cy5标记,将所有的实验样品等量混合而组成内标,用Cy2标记,上样于同一胶中进行电泳分离,经不同光激发下扫描得到不同样品的蛋白质组图谱。所获得的图谱经DeCyder软件分析。结果:江浙蝮蛇毒与白眉蝮蛇毒共有457个蛋白质表达水平相似;较白眉蝮蛇毒,江浙蝮蛇毒有97个蛋白质表达水平显著较好,另外119个蛋白质表达水平显著较低。结论:江浙蝮蛇毒与白眉蝮蛇毒所含蛋白质组具有较大差异,应用时应加以区别。 Objective: To establish two-dlmensional differential gel electrophoretogram of two different Agkistrodon venom with high resolution capacity and stability and analyze the differential proteome. Methods : Agkistrodon halys Palls venom and Agkistrodon blomhoffii Ussurensis venom were labeled with Cy3 or Cy5 respectively, and mixed on the same 2 - D gel along with a Cy2 - labeled mixture of all samples as an internal standard. The gels were imaged using different emission filter respectively and spot-features were analyzed by DeCyder software. Results :457 proteins were similar in Agkistrodon halys Palls venom and Agkistrodon blomhoffii Ussurensis venom. 97 proteins increased remarkably more than 3 folders up while 119 proteins decreased more than 3 folders down in Agkistrodon halys Palls venom than in Agkistrodon blomhoffii Ussurensis venom. Conclusion: Proteome of the two different Agkistrodon venom has obvious difference.
