摘要
目的对41例人胃癌和17例癌旁组织中的金属蛋白酶MMP-2,MMP-9及金属蛋白酶抑制剂TIMP-1基因表达进行了检测;方法应用免疫组织化学(SP)法和原位杂交(cDNA-mRNA)技术对41例胃癌及癌旁组织的金属蛋白酶及其抑制剂进行检测;结果MMP-2及MMP-9蛋白在胃癌及癌旁组织阳性率分别是73.1%(30/41),65.8%(27/41)和22.5%(4/17),41.1%(7/17)。胃癌及癌旁组织的TIMP-1的表达分别是56.9%(23/41)及34.6%(6/17),且MMP-9/TIMP-1的比值与胃癌的侵袭转移呈正相关。一致性检验显示基因的免疫组化和原位杂交检测结果的关系密切(P<0.05);结论MMPS基因的过度表达参与了胃癌的发生发展过程,同时证明TIMP基因的表达参与了肿瘤细胞的生物学行为的改变过程,MMPs/TIMP基因的平衡表达及免疫组化和原位杂交是检测胃癌组织侵袭,转移的可靠方法,本实验的结果为建立肿瘤转移的分子标志提供了理论和实验依据。
Purpose To explove the change of expression of metalloproteinase-2/-9 and tissueinhibitor of metalloproteinase-1 in human gastric carcinoma. Methods lmmunohistochemitry (SPmethod) and in situ hybridization (cDNA-mRNA) were used to detect MMP-2/-9 and TIMP-1 gene ex- pression in tissue of human gastric carcinoma (HGC 41 cases) and near non-tumourous gastric (NT 17cases). Results The positive rates of MMP-2/-9 were 73. 1% (30/41), 65. 8% (27/41 ) and 22. 5% (4/17), 41. 1 % (7/17) in HGC and in NT. The positive rates of TIMP-1 was 56. 9 % (23/41) and 34. 6%(6/17) respectively. The expression of MMP-2/-9 and TIMP-1 at protein level was signficantly correla- tive with that at mRNA level. Conclusion These observations suggest a close association of MMP-2/-9 and TIMP-1 gene expressions with gastricinogenesis. Immunotistochehistry (SP method ) on sectionof paraffin embedded tissue is a reliable method for detecting MMP-2/-9 and TIMP-1 gene expression inHGC.[
