缺氧促进视网膜微血管内皮细胞的增殖及C-fos癌基因表达

摘要 目的研究缺氧在视网膜新生血管形成中的作用。方法采用细胞计数及免疫组化的方法,将培养的视网膜微血管内皮细胞接种于24孔板,分别放入正常及缺氧(50ml·L-1O2)环境中,37℃进行培养。不同时间后进行计数及抗FOS蛋白抗体染色。结果缺氧组的细胞计数均高于正常组(P<0.01),抗FOS蛋白抗体染色,缺氧组明显强于正常组。结论缺氧可以明显促进视网膜微血管内皮细胞的增殖.其机理是通过诱导C-fos癌基因的表达而起作用。 Objective To study the effect of hypoxia on the for mation of pre-retinal neovascularization.Methods The bovine retinal capillary endothelial cells were passaged into 24-well dishs,and the dishs were put into normal and hypoxic chamber respectively. After different times. the cells were counted. At the same time,anti-FOS Protein antibody was used to study the C-fos oncogene expression.Results The cell numbers in the hypoxia group were more than those in the normal group at all time points (P<0. 01),and the staining of anti-FOS protein antibody was strouger than normal.Conclusion Hypoxia can stimulate the growth of retinal capillary endothelial cells. It may act through inducing C-fos oncogene expression.
