两种级别胶质瘤细胞系代谢基因的差异表达 被引量:1

摘要 目的基因的异常表达和功能调节在胶质瘤的进展过程中扮演着重要的作用。然而,导致胶质瘤由低级向高级进展的基因通路远未阐明。本实验拟从不同级别的胶质瘤细胞系筛选代谢相关的差异表达基因,为进一步研究胶质瘤进展的机制提供实验基础。方法以两株不同级别的胶质瘤细胞系(CHG-5,WHOⅡ级;SHG-44,WHOⅣ级)和全反式维甲酸(all-trans retinoic acid,ATRA)为实验模型,运用cDNA微阵列技术进行研究。通过芯片杂交,分别检测CHG-5以及全反式维甲酸诱导前后SHG-44的基因表达谱,筛选差异表达基因;随机选择数个差异基因进行Northern杂交实验,以验证cDNA微阵列的结果。结果本实验发现13个与胶质瘤进展密切相关的代谢基因存在差异表达,它们涉及细胞的糖类和核苷酸代谢。其中,TPI1,BPGM,ALDOA,AKR1B1,LDHA,ADSL,RRM1和NP表达上调,而UGT2B7,ACAT1,IMPDH2,PRPS1和PKM2表达下调。此外,有5个基因(TPI1,BPGM,ALDOA,LDHA和RRM1)在全反式维甲酸诱导的SHG-44细胞中也出现差异表达,提示这些基因不仅存在于不同级别的胶质瘤细胞系之间、而且存在于分化诱导处理的胶质瘤细胞之间。部分差异表达基因为Northern杂交实验所验证。结论利用cDNA微阵列技术首次发现有13个差异表达基因与胶质瘤细胞的代谢密切相关,它们可能在胶质瘤的恶性转化和进展中扮演着重要的作用。
