基因重组人骨形成蛋白-2与骨髓复合移植成骨的观察

摘要 目的:观察基因重组人骨形成蛋白-2(recombinant human bone morphogenetic protein-2,rh-BMP2)与新鲜骨髓复合移植后的成骨能力。方法:从兔股骨抽取骨髓,实验分为3组,第1组在兔股部肌袋中植入自体新鲜骨髓0.3 mL,第2组植入rhBMP2 1 mg,第3组植入骨髓0.3 mL及rhBMP2 1 mg。术后3周取材,定量观察新骨的形成量,并通过组织学观察新骨的形成情况。同时检测新生骨组织的碱性磷酸酶活性、抗压强度等成骨质量。结果:不同材料植入3周后局部均有新骨形成,而骨髓与rhBMP2复合后植入,新骨形成量明显增加,且两者之间有协同作用。组织学观察结果表明:骨髓复合rhBMP2后植入组,新骨更为成熟,碱性磷酸酶活性更高,新生组织的抗压强度更大。结论:rhBMP2和自体新鲜骨髓复合移植后,二者具有协同的成骨作用。 AIM: To investigate bone formation induced by recombinant human bone morphogenetic protein -2(rhBMP2) combined with fresh bone marrow(BM). METHODS: Fresh BM was obtained from rabbit tibia. 0.3 mL BM, 1 mg rhBMP2 and 0.3 mL BM combined with 1 mg rhBMP2 were implanted into rabbit muscle pouch respectively. New bone formation was observed three weeks after operation. RESULTS: Three weeks after implantation of 0.3mL fresh autologus BM, 1 mg rhBMP2, and BM combined with rhBMP2, new bone could be observed both by gross inspection and histologic observation. In the combined group, the amount of new bone was significantly larger than that of the other two groups. Which were demonstrated both by statistics analysis and histologic observation. CONCLUSION: rhBMP2 and BM have synergistic action in the process of bone formation.
