头孢地秦对细胞因子及细胞因子mRNA表达的影响 被引量:2

摘要 目的:探讨头孢地秦对正常及金黄色葡萄球菌感染的败血症小鼠腹腔巨噬细胞(PM)肿瘤坏死因子α(TNFα)与白细胞介素6(IL6)分泌及其mRNA表达的影响。方法:采集小鼠PM,体外给药,分离细胞培养上清。生物检测法测定TNFα,ELISA法测定IL6;RTPCR方法检测mRNA的表达。结果:头孢地秦使正常小鼠PM培养上清中TNFα的活性降低,使金黄色葡萄球菌感染小鼠PM48h培养上清中TNFα活性升高。200mg/L的头孢地秦对正常小鼠PM的TNFαmRNA表达有显著抑制作用,但对感染小鼠TNFαmRNA表达没有明显影响。头孢地秦抑制正常小鼠PM48h培养上清中IL6的产生,明显增加金黄色葡萄球菌感染小鼠PM培养上清中的IL6。头孢地秦对IL6mRNA表达的影响与细胞因子的改变一致。结论:头孢地秦对正常和感染状态下小鼠TNFα和IL6产生的影响不同。 ABSTRACT Objective: To investigate the effects of cefodizime (CDZM) on production of TNFα, IL-6 and their mRNA expression in murine peritoneal macrophage (PM) from normal and Staphylococcus aureus infected mouse. Methods: Mouse PM were treated in vitro with cefodizime, cell culture supernatant was separated. TNFα was detected by biological assay, IL6 by ELISA, and the level of mRNA with the method of reverse transcriptionpolymerase chain reaction (RTPCR). Results:TNFα production was decreased in the supernatant of normal murine PM while increased in the supernatant of Staphylococcus aureus infected murine PM by the effect of CDZM. The maximal effect was observed after 48 h of incubation. CDZM significantly restrained TNFα mRNA expression in normal murine PM at dose of 200 mg/L. Its promoting effect to TNFα mRNA expression had no statistical difference to the basal level. CDZM slightly inhibited the production of IL-6 in the supernatant of normal murine PM after 48 h incubation, but significantly stimulated the production of IL-6 in Staphylococcus aureus infected murine PM. Coordinate result was obtained in the expression of IL-6 mRNA. Conclusion: The effects of CDZM on TNFα and IL-6 production are different between normal and Staphylococcus aureus infected mouse.The corresponding mRNA expression are coordinately different.
