摘要
应用肌酸比色法快速测定发酵液中的3-羟基丁酮含量,考察了3-羟基丁酮发酵体系中系列副产物对该方法测定结果的影响。发现除3-羟基丁酮的还原态副产物2,3-丁二醇对测定结果有微弱影响外,其余副产物(乙醇、乙酸、乳酸和丁二酸)以及培养基对测定结果均没有影响;进一步将该方法与气相色谱法的检测结果进行了对比分析,表明该法测定3-羟基丁酮含量相对于气相色谱法测定结果误差在3%以内,平均回收率为98.90%,相对标准偏差(RSD)为0.25%;利用该法可以快速有效测定发酵液中3-羟基丁酮的含量并能够满足高产菌株选育过程中定性与定量检测的需要。
A method using creatinine colorimetric assay was developed to determine the content of acetoin in us subtilis fermentation broth quickly. The effect of main byproduct 2,3-butanediol in the acetoin fermentation broth on the coloration result was investigated, the result showed that acetoin concentration measured by the colorimetric method was in good agreement with that by GC method within 3% deviation. The average recovery rate was 98.90%, the relative standard deviation (RSD) was 0.25%. The conclusion was that the creatinine colorimetric assay method could be applied to determine the content of acetoin in the fermentation broth, and it could also meet the need of the qualitative and quantitative analysis of acetoin for breeding high acetoin producing strain.
