中药筋脉通对高糖培养雪旺细胞增殖及NGF表达的影响 被引量:29

摘要 目的:从细胞生物学水平观察筋脉通含药血清对高糖培养雪旺细胞(SC)的增殖及其分泌神经生长因子(NGF)的影响。方法:体外培养大鼠SC,采用血清药理学方法制备筋脉通含药血清,观察筋脉通含药血清对高糖培养SC增殖活性的影响,检测筋脉通含药血清对高糖培养SC分泌NGF的影响。结果:高糖培养条件下,SC增殖及分泌NGF的能力明显减弱,筋脉通含药血清可增强高糖培养SC的增殖能力,上调NGF的表达水平,作用与神经妥乐平含药血清相似。结论:中药筋脉通可有效促进高糖环境培养的SC的增殖能力及其分泌NGF的水平。 Objective : To investigate the effects of medicated serum of Jinmaitong Capsule(JMT) on proliferation and expression of nerve growth factor ( NGF ) in schwann cells ( SC ) cultured in high glucose. Methods: SC derivedfrom postnatal rats was cultured in vitro. The serum pharmacological method was applied to prepare medicated serum of Jinmaitong Capsule and then the serum was added to SC cultured in vitro. Effects of medicated serum of JMT on proliferation of SC cultured in high glucose were detected by using methyl thiazolyl tetrazolium (MTT) assay. Furthermore, confocal laser scanning microscope was deployed to determine NGF levels in SC cultured in high glucose. Results: High glucose inhibited the proliferation and expression of NGF of SC remarkably. The medicated serum of JMT could promote proliferation and expression of NGF effectively in SC cultured in high glucose and its role was similar to that of Neurotropin. Conclusion: Traditional Chinese medicine JMT can promote proliferation and expression of NGF in SC cultured in high glucose significantly.
