溶菌酶标准酶活曲线测定方法的改进 被引量:2

摘要 天然溶菌酶活性(标准酶活)的测定是蛋白质复性研究过程中的一个关键步骤。为了增加测定数据的准确性和可比性,改进了测定标准酶活的方法。在固定其他条件的前提下,对于影响标准酶活的重要因素-温度,采用空调控制室温的方法,使其规律性由低到高变化。同时将测活液放置在与所控制室温相同的水浴环境中保持温度,测出各种温度下曲线的斜率,用直线进行拟合。对于其他没有测定的温度点,可以从拟合曲线中求出。这样,通过一组实验便可以得到任意温度下的标准酶活。 The measurement of the natural activity of lysozyme is a committed step in the protein renaturation researching. In order to improve the veracity and the comparability, the method of measuring the standard activity of lysozyme was improved. With the other condition invariable, the air-conditioning was used to modulate the temperature, which is an important factor to influence the standard activity of lysozyme. In the test, the temperature should be changed from low to high regularly. At the same time ,the determining activity liquid should be placed in the water-bath to keep its temperature the same as the room' s. Then we got the slope of the curve in different temperatures, and had the straight line fitted. For the temperature which hadn' t been measured, it could be solved by the fitted curve. In this method the standard activity of lysozyme of every temperature could be found only by doing a set of experiment.
