人参皂苷Rb1抗SD大鼠海马神经元的缺氧损伤作用 被引量:7

摘要 目的探讨人参皂苷Rb1对脑缺氧状态下SD大鼠海马神经元的保护作用。方法以19d的SD胎鼠脑作为取材对象,经过条件培养基纯化培养,建立海马神经元原代培养体系;选用人参皂苷Rb1作为干预药物,通过MTT法观察缺氧环境对海马神经元生长活力的影响;通过免疫荧光细胞化学法检测胞内caspase-3活性表达;通过TUNEL法观察缺氧环境对海马神经元凋亡的影响;通过化学发光法检测胞内ATP的表达水平。结果与缺氧组相比,Rb1提高了经缺氧损害致凋亡的海马神经元生长增殖活力;对缺氧诱导的海马神经元caspase-3活性增高有明显的抑制作用;还可减轻因缺氧降低的胞内ATP水平。结论人参皂苷Rb1能够减轻缺氧诱导的细胞损害和海马神经元损伤程度。 Objective To explore the protective effect of ginsenoside Rbl on hippocampal neurons of hypoxic rats. Methods The primary culture system of hippocampal neurons was established with 19 d SD embryo rat brain through the condition culture medium. Then the neuron' s proliferation and apoptosis, caspase-3 activity and ATP level were detected under the condition of hypoxia by MTT, TUNEL,immunofluorescence and chemiluminescence,with the ginsenoside Rbl as an intervention drug. Results After the hippocampal neurons of hypoxic rats were intervented by ginsenoside Rbl, the neuronal proliferation vigor increased, the apoptosis was suppressed, and caspase-3 activity and ATP production increased. Conclusion Ginsenoside Rbl could reduce hypoxia-induced cell injury and damage of hippocampal neurons.
